    Period and toroidal knot mosaics

    Knot mosaic theory was introduced by Lomonaco and Kauffman in the paper on `Quantum knots and mosaics' to give a precise and workable definition of quantum knots, intended to represent an actual physical quantum system. A knot (m,n)-mosaic is an m ⁣× ⁣nm \! \times \! n matrix whose entries are eleven mosaic tiles, representing a knot or a link by adjoining properly. In this paper we introduce two variants of knot mosaics: period knot mosaics and toroidal knot mosaics, which are common features in physics and mathematics. We present an algorithm producing the exact enumeration of period knot (m,n)-mosaics for any positive integers m and n, toroidal knot (m,n)-mosaics for co-prime integers m and n, and furthermore toroidal knot (p,p)-mosaics for a prime number p.     Transient receptor potential canonical type 3 channels control the vascular contractility of mouse mesenteric arteries

    Transient receptor potential canonical type 3 (TRPC3) channels are non-selective cation channels and regulate intracellular Ca2+ concentration. We examined the role of TRPC3 channels in agonist-, membrane depolarization (high K+)-, and mechanical (pressure)-induced vasoconstriction and vasorelaxation in mouse mesenteric arteries. Vasoconstriction and vasorelaxation of endothelial cells intact mesenteric arteries were measured in TRPC3 wild-type (WT) and knockout (KO) mice. Calcium concentration ([Ca2+]) was measured in isolated arteries from TRPC3 WT and KO mice as well as in the mouse endothelial cell line bEnd.3. Nitric oxide (NO) production and nitrate/nitrite concentrations were also measured in TRPC3 WT and KO mice. Phenylephrine-induced vasoconstriction was reduced in TRPC3 KO mice when compared to that of WT mice, but neither high K+- nor pressure-induced vasoconstriction was altered in TRPC3 KO mice. Acetylcholine-induced vasorelaxation was inhibited in TRPC3 KO mice and by the selective TRPC3 blocker pyrazole-3. Acetylcholine blocked the phenylephrine-induced increase in Ca2+ ratio and then relaxation in TRPC3 WT mice but had little effect on those outcomes in KO mice. Acetylcholine evoked a Ca2+ increase in endothelial cells, which was inhibited by pyrazole-3. Acetylcholine induced increased NO release in TRPC3 WT mice, but not in KO mice. Acetylcholine also increased the nitrate/nitrite concentration in TRPC3 WT mice, but not in KO mice. The present study directly demonstrated that the TRPC3 channel is involved in agonist-induced vasoconstriction and plays important role in NO-mediated vasorelaxation of intact mesenteric arteries.Fil: Yeon, Soo-In.     The Benefit of Hydroxyapatite-augmented Pedicle Screw Fixation in Degenerative Spondylolisthesis Patients with Osteoporosis

    Objective A key determinant of the performance of pedicle screw systems is the strength of their attachment to the spine. The authors initially injected hydroxyapatite cement into the prepared screw hole before actual screw insertion in order to provide additional strength to this system and assessed its benefit in the fusion operation for the patients with osteoporosis. Methods Among the 21 patients who were diagnosed as grade I degenerative spondylolisthesis on L4-5 level with osteoporosis, nine patients were treated by transforaminal lumbar interbody fusion (TLIF) with screw augmentation with hydroxyapatite and the rest 12 patients underwent TLIF only. The plain radiographs were periodically checked during and up to 2 years after the operation. The changes of the angles between the vertebral bodies, between the upper and lower screws, between the screw and lower endplate of each body, and posterior disc height changes were serially measured and compared between these two groups. Results The angle between the upper screw and the lower endplate of upper vertebral body showed gradual decrease from 8.93° to 5.79° for the TLIF only group while well maintained for the augmentation group from 6.84° to 6.93° (p=0.053). Most of these significant angle changes occurred between 1 month and 6 months after the operation (p<0.05). Conclusion Most of the screw construct changes occurred in the upper level of fixation during 1 month to 6 months after fusion surgery in spondylolisthesis patients with osteoporosis. For these, the screw augmentation with hydroxyapatite demonstrated the improvement in maintaining the screw construct especially in the upper level of fixation

    Human endometrial cell coculture reduces the endocrine disruptor toxicity on mouse embryo development

    BACKGROUNDS: Previous studies suggested that endocrine disruptors (ED) are toxic on preimplantation embryos and inhibit development of embryos in vitro culture. However, information about the toxicity of endocrine disruptors on preimplantation development of embryo in human reproductive environment is lacking. METHODS: Bisphenol A (BPA) and Aroclor 1254 (polychlorinated biphenyls) were used as endocrine disruptors in this study. Mouse 2-cell embryos were cultured in medium alone or vehicle or co-cultured with human endometrial epithelial layers in increasing ED concentrations. RESULTS: At 72 hours the percentage of normal blastocyst were decreased by ED in a dose-dependent manner while the co-culture system significantly enhanced the rate and reduced the toxicity of endocrine disruptors on the embryonic development in vitro. CONCLUSIONS: In conclusion, although EDs have the toxic effect on embryo development, the co-culture with human endometrial cell reduced the preimplantation embryo from it thereby making human reproductive environment protective to preimplantation embryo from the toxicity of endocrine disruptors

    Structural Optimization of a Knuckle with Consideration of Stiffness and Durability Requirements

    The automobile’s knuckle is connected to the parts of the steering system and the suspension system and it is used for adjusting the direction of a rotation through its attachment to the wheel. This study changes the existing material made of GCD45 to Al6082M and recommends the lightweight design of the knuckle as the optimal design technique to be installed in small cars. Six shape design variables were selected for the optimization of the knuckle and the criteria relevant to stiffness and durability were considered as the design requirements during the optimization process. The metamodel-based optimization method that uses the kriging interpolation method as the optimization technique was applied. The result shows that all constraints for stiffness and durability are satisfied using A16082M, while reducing the weight of the knuckle by 60% compared to that of the existing GCD450

    GBParsy: A GenBank flatfile parser library with high speed

    <p>Abstract</p> <p>Background</p> <p>GenBank flatfile (GBF) format is one of the most popular sequence file formats because of its detailed sequence features and ease of readability. To use the data in the file by a computer, a parsing process is required and is performed according to a given grammar for the sequence and the description in a GBF. Currently, several parser libraries for the GBF have been developed. However, with the accumulation of DNA sequence information from eukaryotic chromosomes, parsing a eukaryotic genome sequence with these libraries inevitably takes a long time, due to the large GBF file and its correspondingly large genomic nucleotide sequence and related feature information. Thus, there is significant need to develop a parsing program with high speed and efficient use of system memory.</p> <p>Results</p> <p>We developed a library, GBParsy, which was C language-based and parses GBF files. The parsing speed was maximized by using content-specified functions in place of regular expressions that are flexible but slow. In addition, we optimized an algorithm related to memory usage so that it also increased parsing performance and efficiency of memory usage. GBParsy is at least 5 - 100× faster than current parsers in benchmark tests.</p> <p>Conclusion</p> <p>GBParsy is estimated to extract annotated information from almost 100 Mb of a GenBank flatfile for chromosomal sequence information within a second. Thus, it should be used for a variety of applications such as on-time visualization of a genome at a web site.</p

    Molecular cloning and biochemical characterization of a novel erythrose reductase from Candida magnoliae JH110

    <p>Abstract</p> <p>Background</p> <p>Erythrose reductase (ER) catalyzes the final step of erythritol production, which is reducing erythrose to erythritol using NAD(P)H as a cofactor. ER has gained interest because of its importance in the production of erythritol, which has extremely low digestibility and approved safety for diabetics. Although ERs were purified and characterized from microbial sources, the entire primary structure and the corresponding DNA for ER still remain unknown in most of erythritol-producing yeasts. <it>Candida magnoliae </it>JH110 isolated from honeycombs produces a significant amount of erythritol, suggesting the presence of erythrose metabolizing enzymes. Here we provide the genetic sequence and functional characteristics of a novel NADPH-dependent ER from <it>C. magnoliae </it>JH110.</p> <p>Results</p> <p>The gene encoding a novel ER was isolated from an osmophilic yeast <it>C. magnoliae </it>JH110. The ER gene composed of 849 nucleotides encodes a polypeptide with a calculated molecular mass of 31.4 kDa. The deduced amino acid sequence of ER showed a high degree of similarity to other members of the aldo-keto reductase superfamily including three ER isozymes from <it>Trichosporonoides megachiliensis </it>SNG-42. The intact coding region of ER from <it>C. magnoliae </it>JH110 was cloned, functionally expressed in <it>Escherichia coli </it>using a combined approach of gene fusion and molecular chaperone co-expression, and subsequently purified to homogeneity. The enzyme displayed a temperature and pH optimum at 42°C and 5.5, respectively. Among various aldoses, the <it>C. magnoliae </it>JH110 ER showed high specific activity for reduction of erythrose to the corresponding alcohol, erythritol. To explore the molecular basis of the catalysis of erythrose reduction with NADPH, homology structural modeling was performed. The result suggested that NADPH binding partners are completely conserved in the <it>C. magnoliae </it>JH110 ER. Furthermore, NADPH interacts with the side chains Lys252, Thr255, and Arg258, which could account for the enzyme's absolute requirement of NADPH over NADH.</p> <p>Conclusions</p> <p>A novel ER enzyme and its corresponding gene were isolated from <it>C. magnoliae </it>JH110. The <it>C. magnoliae </it>JH110 ER with high activity and catalytic efficiency would be very useful for <it>in vitro </it>erythritol production and could be applied for the production of erythritol in other microorganisms, which do not produce erythritol.</p

    Distribution and Kinematics of H I through Raman He II Spectroscopy of NGC 6302

    The young planetary nebula NGC 6302 is known to exhibit Raman-scattered He II features at 6545 and 4851 Angstrom. These features are formed through inelastic scattering of He IIλλ\lambda\lambda 1025 and 972 with hydrogen atoms in the ground state, for which the cross sections are 1.2×10211.2 \times 10^{-21} and 1.4×1022 cm21.4\times 10^{-22} {\rm\ cm^2}, respectively. We investigate the spectrum of NGC 6302 archived in the ESO Science Portal. Our Gaussian line fitting analysis shows that the Raman-scattered He II features are broader and more redshifted than the hypothetical model Raman features that would be formed in a cold static H I medium. We adopt a simple scattering geometry consisting of a compact He II emission region surrounded by a H I medium to perform Monte Carlo simulations using the radiative transfer code STaRS{\it STaRS}. Our simulations show that the H I region is characterized by the H I column density NHI=3×1021 cm2N_{\rm HI}=3\times 10^{21}{\rm\ cm^{-2}} with the random speed component vran=10 km s1v_{\rm ran}=10{\rm\ km\ s^{-1}} expanding with a speed $v_{\rm exp}= 13{\rm\ km\ s^{-1}}fromtheHeIIemissionregion.Basedonourbestfitparameters,weestimatetheHImassoftheneutralmedium from the He II emission region. Based on our best fit parameters, we estimate the H I mass of the neutral medium M_{\rm HI} \simeq 1.0\times 10^{-2}\ {\rm M_\odot}$, pointing out the usefulness of Raman He II spectroscopy as a tool to trace H I components.Comment: 12 pages, 8 figures, accepted for publication in Ap

    Exploration of New Electroacupuncture Needle Material

    Background. Electro Acupuncture (EA) uses the acupuncture needle as an electrode to apply low-frequency stimulation. For its safe operation, it is essential to prevent any corrosion of the acupuncture needle. Objective. The aim of this study is to find an available material and determine the possibility of producing a standard EA needle that is biocompatible. Methods. Biocompatibility was tested by an MTT assay and cytotoxicity testing. Corrosion was observed with a scanning electron microscope (SEM) after 0.5 mA, 60 min stimulation. The straightness was measured using a gap length of 100 mm, and tensile testing was performed by imposing a maximum tensile load. Results. Phosphor bronze, Ni coated SS304, were deemed inappropriate materials because of mild-to-moderate cytotoxicity and corrosion. Ti-6Al-4V and SS316 showed no cytotoxicity or corrosion. Ti-6Al-4V has a 70 times higher cost and 2.5 times lower conductivity than SS316. The results of both straightness and tensile testing confirmed that SS316 can be manufactured as a standard product. Conclusion. As a result, we confirmed that SS316 can be used a new EA electrode material. We hope that a further study of the maximum capacity of low-frequency stimulation using an SS316 for safe operation
